What's the difference between brightness and contrast?

Brightness adds or subtracts a fixed value from each pixel, uniformly lightening or darkening the image. Contrast, on the other hand, spreads apart or brings together the light and dark values, making the image look more or less "sharp" visually.

Can I combine both adjustments?

Yes, both adjustments are applied one after the other (brightness first, then contrast) and can be combined freely to achieve the desired result.
